Reconstructed Human Epidermis: an Efficient Prediction Tool

چکیده
RHE has the ultimate advantage to grow donor epidermal cells in a serumfree culture environment: after rapidly proliferating preparative keratinocyte cultures have been obtained, the yielded epidermal cells are seeded on inert filter substrates that are lifted to the air-liquid interface in a humidified-air incubator. A fully-defined nutrient medium feeds the basal cells through the filter substratum. After 14 days, a stratified epidermis has formed that closely resembles human epidermis in vivo. Morphologically, these cultures exhibit a well-stratified epithelium and cornified epidermis with significantly improved barrier function and metabolic activity. Differentiation markers such as suprabasal keratins, integrin b4, integrin a6, fibronectin, involucrin, filaggrin, trichohyalin, type I, III, IV, V and VII collagen, laminin, heparan sulfate and membrane-bound transglutaminase have been found to be expressed similar to those of the epidermis. Esterase Activity and Intracellular Localization in Reconstructed Human Epidermal Cultured Skin Models
BACKGROUND Reconstructed human epidermal culture skin models have been developed for cosmetic and pharmaceutical research. OBJECTIVE This study evaluated the total and carboxyl esterase activities (i.e., Km and Vmax , respectively) and localization in two reconstructed human epidermal culture skin models (LabCyte EPI-MODEL [Japan Tissue Engineering] and EpiDerm [MatTek/Kurabo]). The usefulnes...
متن کاملThe EPISKIN Phototoxicity Assay (EPA): Development of an in vitro tiered strategy to predict phototoxic potential
The aim of the study was to investigate the ability of human reconstructed epidermis EpiSkin to assess the phototoxic potential of chemicals following topical or systemic treatment. Three categories of chemicals according to their available human phototoxic potential were evaluated: systemic phototoxic, topical phototoxic and non phototoxic chemicals. Products were applied topically or added to...
متن کاملModel of human epidermis reconstructed in vitro with keratinocytes and melanocytes on dead de-epidermized human dermis.
CONTEXT Recent progress in the field of epithelial culture techniques has allowed the development of culture systems in which the reconstructed epidermis presents characteristics of morphological differentiation similar to those seen in vivo. Human epidermis reconstructed in vitro may be used as the best alternative for the in vitro testing of the toxicology and efficiency of products for topic...
متن کاملModel for human skin reconstructed in vitro composed of associated dermis and epidermis.
CONTEXT AND OBJECTIVE The technique of obtaining human skin with dermis and epidermis reconstructed from cells isolated from patients can enable autologous skin grafting on patients with few donor sites. It also enables in vitro trials on chemicals and drugs. The objective of this work was to demonstrate a method for obtaining human skin composed of associated dermis and epidermis, reconstructe...
متن کاملCharacterization and comparison of reconstructed skin models: morphological and immunohistochemical evaluation.
Reconstructed human skin equivalents are currently being investigated as in vitro models for the prediction of human skin toxicity and irritation responses. Three different industrial reconstructed skin models (EpiDerm, Episkin and SkinEthic) and one in-house equivalent were characterized and compared using light microscopy, immunohistochemistry and reduction of (3-[4,5-dimethylthiazole-2-yl]-2...
